Protein Function
Heterogeneous nuclear ribonucleoprotein (hnRNP) that associates with nascent pre-mRNAs, packaging them into hnRNP particles. The hnRNP particle arrangement on nascent hnRNA is non- random and sequence-dependent and serves to condense and stabilize the transcripts and minimize tangling and knotting. Packaging plays a role in various processes such as transcription, pre-mRNA processing, RNA nuclear export, subcellular location, mRNA translation and stability of mature mRNAs (PubMed:19099192). Forms hnRNP particles with at least 20 other different hnRNP and heterogeneous nuclear RNA in the nucleus. Involved in transport of specific mRNAs to the cytoplasm in oligodendrocytes and neurons: acts by specifically recognizing and binding the A2RE (21 nucleotide hnRNP A2 response element) or the A2RE11 (derivative 11 nucleotide oligonucleotide) sequence motifs present on some mRNAs, and promotes their transport to the cytoplasm (PubMed:10567417). Specifically binds single-stranded telomeric DNA sequences, protecting telomeric DNA repeat against endonuclease digestion (By similarity). Also binds other RNA molecules, such as primary miRNA (pri-miRNAs): acts as a nuclear 'reader' of the N6-methyladenosine (m6A) mark by specifically recognizing and binding a subset of nuclear m6A-containing pri-miRNAs. Binding to m6A-containing pri- miRNAs promotes pri-miRNA processing by enhancing binding of DGCR8 to pri-miRNA transcripts (PubMed:26321680). Involved in miRNA sorting into exosomes following sumoylation, possibly by binding (m6A)-containing pre-miRNAs (PubMed:24356509). Acts as a regulator of efficiency of mRNA splicing, possibly by binding to m6A- containing pre-mRNAs (PubMed:26321680).

Western Blot (WB) of anti-TP53BP1 antibody

Figure 1. Western blot analysis of TP53BP1 using anti-TP53BP1 antibody (MBS1758071).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: human Hela whole cell lysates,
Lane 2: rat heart tissue lysates,
Lane 3: mouse heart tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-TP53BP1 antigen affinity purified polyclonal antibody (#MBS1758071) at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (#MBS176460) with Tanon 5200 system. A specific band was detected for TP53BP1 at approximately 300-350 kDa. The expected band size for TP53BP1 is at 210 kDa.

Figure 2. IF analysis of TP53BP1 using anti-TP53BP1 antibody (MBS1758071) and anti-Beta Tubulin antibody (M01857-3).
TP53BP1 was detected in immunocytochemical section of U2OS cell. Enzyme antigen retrieval was performed using IHC enzyme antigen retrieval reagent (#MBS176582) for 15 mins. The cells were blocked with 10% goat serum. And then incubated with 5ug/mL rabbit anti-TP53BP1 Antibody (MBS1758071) and mouse anti-Beta Tubulin antibody (M01857-3) overnight at 4 degree C. Cy3 Conjugated Goat Anti-Rabbit IgG (BA1032) and DyLight488 Conjugated Goat Anti-Mouse IgG (BA1126) were used as secondary antibody at 1:500 dilution and incubated for 30 minutes at 37 degree C. Visualize using a fluorescence microscope and filter sets appropriate for the label used.

Flow Cytometry (FC/FACS) of anti-TP53BP1 antibody

Figure 3. Flow Cytometry analysis of HepG2 cells using anti-TP53BP1 antibody (MBS1758071).
Overlay histogram showing HepG2 cells stained with MBS1758071 (Blue line). To facilitate intracellular staining, cells were fixed with 4% paraformaldehyde and permeabilized with permeabilization buffer. The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-TP53BP1 Antibody (MBS1758071, 1ug/1x106 cells) for 30 min at 20 degree C. DyLight488 conjugated goat anti-rabbit IgG (MBS177405, 5-10ug/1x106 cells) was used as secondary antibody for 30 minutes at 20 degree C. Isotype control antibody (Green line) was rabbit IgG (1ug/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.

UniProt Comments for TP53BP1

53BP1: a DNA damage checkpoint protein that interacts with HDAC4 protein to mediate the DNA damage response. Inhibits E2F1-mediated apoptosis in prostate cancer cells. Recruited to double strand breaks via the binding of its Tudor domain to dimethylated H3 K20. Asymmetrically dimethylated on Arg residues by PRMT1. Methylation is required for DNA binding. Binds to the central domain of p53. Interacts with Artemis. Interacts with histone H2AFX and this requires phosphorylation of H2AFX on S139. Interacts with histone H4 that has been dimethylated at K20. Has low affinity for histone H4 containing monomethylated K20. Does not bind histone H4 containing unmethylated or trimethylated K20. Has low affinity for histone H3 that has been dimethylated on K79. Does not bind unmethylated histone H3. Phosphorylated at basal level in the absence of DNA damage. Hyper-phosphorylated in an ATM-dependent manner in response to DNA damage induced by ionizing radiation. Hyper-phosphorylated in an ATR-dependent manner in response to DNA damage induced by UV irradiation. Two alternatively spliced human isoforms have been observed.
