Western Blot (WB) of anti-OCLN antibody

Figure 1. Western blot analysis of Occludin/Ocln using anti-Occludin/Ocln antibody (MBS1758166).
Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel)/90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 30 ug of sample under reducing conditions.
Lane 1: rat liver tissue lysates,
Lane 2: rat RH35 whole cell lysates,
Lane 3: mouse liver tissue lysates,
Lane 4: mouse kidney tissue lysates.
After electrophoresis, proteins were transferred to a nitrocellulose membrane at 150 mA for 50-90 minutes. Blocked the membrane with 5% non-fat milk/TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-Occludin/Ocln antigen affinity purified polyclonal antibody (#MBS1758166) at 0.5ug/mL overnight at 4 degree C, then washed with TBS-0.1%Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:5000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit (#MBS176460) with Tanon 5200 system. A specific band was detected for Occludin/Ocln at approximately 59 kDa. The expected band size for Occludin/Ocln is at 59 kDa.

UniProt Comments for OCLN

occludin: May play a role in the formation and regulation of the tight junction (TJ) paracellular permeability barrier. It is able to induce adhesion when expressed in cells lacking tight junctions. Interacts with TJP1/ZO1 and with VAPA. Localized at tight junctions of both epithelial and endothelial cells. Highly expressed in kidney. Not detected in testis. Belongs to the ELL/occludin family. 7 isoforms of the human protein are produced by alternative splicing.

Protein type: Membrane protein, multi-pass; Membrane protein, integral; Cell adhesion; Motility/polarity/chemotaxis

Cellular Component: apical plasma membrane; apicolateral plasma membrane; cell junction; cell surface; cytoplasmic vesicle; cytosol; endocytic vesicle; integral to membrane; intercellular junction; membrane; plasma membrane; tight junction
