Principle of the Assay||The Rat IGFBP5 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Rat IGFBP5 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Rat IGFBP5 coated on a 96-well plate. Standards and samples are pipetted into the wells and IGFBP5 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Rat IGFBP5 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IGFBP5 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Insulin-like growth factor-binding protein 5 is a protein that in humans is encoded by the IGFBP5 gene. The IGFBP5 gene was mapped to chromosome 2q33-q34. The expression of IGFBP5 by stable transfection and adenovirus-mediated infection was inhibitory to growth in 2 human breast cancer cell lines. Stable expression of IGFBP5 in the breast cancer cell lines also inhibited the formation and growth of tumors following injection in athymic mice. IGFBP5 is believed a growth inhibitor and proapoptotic agent in breast cancer cells.
