Profilin (PFN1) Antibody – Rabbit Polyclonal

For Research Use Only. Not for use in diagnostic procedures. Catalog #: MBS474133

Profilins are small actin-binding proteins that have functions in cell motility, cytokinesis, gene transcription, endocytosis and neuronal plasticity. Four profilin isoforms have been identified in mammals. Profilin-1 (PFN1) and profilin-2a (PFN2a) isoforms… More details

Purity/Purification
Affinity Purified
Specificity
This antibody was affinity purified using Profilin (a.a. 126-137) peptide (without carrier). The antibody detects a 15 kDa band corresponding to the apparent molecular mass of Profilin-1 in human umbilical vein endothelial cells (HUVEC) and Jurkat cells.
Preparation and Storage
Store at -20 degree C. Stable for 1 year.
Molecular Weight
15 kDa
OMIM
176610
3D Structure
ModBase 3D Structure for P07737
Immunogen
Profilin synthetic peptide (coupled to carrier protein) that includes amino acids 126 to 137 in human Profilin-1. This sequence is well conserved in rat and mouse Profilin-1, has some homology to Profilin-2a and Profilin-2b, and low homology to Profiin-3 and Profilin-4.
Applications Tested/Suitable for anti-PFN1 antibody
Western Blot (WB), ELISA (EIA)
Application Notes for anti-PFN1 antibody
ELISA: 1:2000
WB: 1:1000
WB: Western blot membranes were incubated with diluted antibody in 5% non-fat milk, PBS, 0.04% Tween20 for 1 hour at room temperature.

Western Blot (WB) of anti-PFN1 antibody

Western blot of HUVEC stimulated with Pervanadate (1 mM) for 30 min (lanes 1-4). The blots were treated with alkaline phosphatase to remove phosphorylation (lanes 2 & 4), then probed with anti-Profilin (a.a. 126-137) (lanes 1 & 2) or anti-Profilin (Tyr-129) phospho-specific (lanes 3 & 4). The antibodies detect profilin at 15 kDa. In lanes 3 & 4, the antibody also detects an unknown 20 kDa protein.

Other Notes

Small volumes of anti-PFN1 antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

NCBI Summary for PFN1

This gene encodes a member of the profilin family of small actin-binding proteins. The encoded protein plays an important role in actin dynamics by regulating actin polymerization in response to extracellular signals. Deletion of this gene is associated with Miller-Dieker syndrome, and the encoded protein may also play a role in Huntington disease. Multiple pseudogenes of this gene are located on chromosome 1.
