Applications are based on unconjugated antibody.

Western Blot (WB) of anti-ATR antibody

Western Blotting Analysis: Representative lot data. UV irradiated HeLa cell lysates (Lanes 1-3) or UV irradiated COS1 (Lanes 4-6) lysates were probed with Anti-phospho-ATR (Ser428) (1:200 dilution). Lane 1 and 4: Antibody +No peptide Lane 2 and 5: Antibody +Non-phospho peptide Lane 3 and 6: Antibody +Phospho peptide Proteins were visualized using a Donkey Anti-Rabbit conjugated to HRP and a chemiluminescence detection system. Arrow indicates ATR phosphorylated at (Ser428) (~260kD).

Other Notes

Small volumes of anti-ATR antibody vial(s) may occasionally become entrapped in the seal of the product vial during shipment and storage. If necessary, briefly centrifuge the vial on a tabletop centrifuge to dislodge any liquid in the container`s cap. Certain products may require to ship with dry ice and additional dry ice fee may apply.

UniProt Comments for ATR

ATR: a ser/thr kinase of the PIKK family most closely related to ATM. Activates checkpoint signaling upon genotoxic stresses such as ionizing radiation (IR), ultraviolet light (UV), or DNA replication stalling, thereby acting as a DNA damage sensor. Recognizes the substrate consensus sequence [ST]Q. Phosphorylates BRCA1, Chk1, MCM2, RAD17, RPA2, SMC1 and p53, which collectively inhibit DNA replication and mitosis and promote DNA repair, recombination and apoptosis. Phosphorylates 'Ser-139' of histone variant H2AX/H2AFX at sites of DNA damage, thereby regulating DNA damage response mechanism. Required for FANCD2 ubiquitination. Critical for maintenance of fragile site stability and efficient regulation of centrosome duplication.
